Disregard positioning (especially on tanks) because that’s just something you can’t control. I can think of is Sally, Ursula and Yzma. Sally because you want to make sure you have poison stacks before activating, and Yzma because you can craft longer potions. Ursula needs good timing as well.

Miguel is much less helpful on Auto. Same with Bo, who can bring the enemy into your backlines.

Olaf is best on manual: if you put him on auto, his white skill will be activated on the wrong time, especially when you are taking down a powerful team. So, putting Olaf on manual is recommended.

Yes but that’s just positioning. A lot of other heroes have that problem.
